中文摘要: 中药复方制剂对人体疾病的调节是药物分子与人体内生物分子间相互作用的综合结果,通过现代的系统生物学技术这一有力手段对其进行研究,有利于阐明复方制剂治疗作用的生化机制和处方配伍的内在规律。代谢组学基于整体性研究和系统性的理念比较适合中医药作用的特点,本项目拟采用代谢组学技术对中药复方双参通冠方的心血管疾病治疗作用进行机理研究。考虑到复方多靶点的特点,应用与其功效相关的两种动物模型- - 血瘀证和心肌缺血动物模型为对象进行研究,以生理学检测指标为基础,以中药的治疗作用和配伍增效为切入点,围绕生物标志物和生物代谢途径的挖掘,进行作用机制和配伍科学性研究。分别从动物模型、复方干预、配伍增效三个层次进行系统的代谢轮廓谱分析,通过分析复方对动物模型内源性成分的影响,建立该复方制剂治疗作用的代谢组学评价方法,通过对比复方与单味药干预作用在代谢组学上的差别来探讨其配伍的科学性。

英文摘要: The regulation of traditional Chinese medicine (TCM) on the human disease is the combined result of the interaction between drug molecules and biological molecules of the human body. Modern systems biology technology is a valuable tool for the research on elucidating the biochemical mechanism of the therapeutic effect and compatibility of TCM formula. Metabonomics based on the integrity of research and systematic concept is more suitable for the characteristics of the role of Chinese medicine. This project intends to adopt the metabonomic study on the therapeutic effect of cardiovascular diseases of TCM "Shuang Shen Tong Guan" (SSTG)formula. As the characteristics of multi-target compound of TCM, the two animal models related to its efficacy - the blood stasis syndrome and myocardial ischemia animal models are selected as the objects of study.Metabonomic profile analysis from three levels of animal models, compounds interference, compatibility efficiency was performed for TCM mechanism study. Through the analysis of endogenous metabolites of animal models, the metabonomic method for evaluating the therapeutic effect of SSTG formula was established.
