中文摘要: 飞蝗受到外界环境种群密度的影响能够在群居型飞蝗和散居型飞蝗之间发生转变,这种两型转变的现象是当今表型可塑性研究领域的热门问题之一。miRNA是一种非编码并且具有功能的RNA分子,能够通过与碱基配对的形式在转录后水平抑制基因表达,实现对多种生物学过程的调节。通过RNAi实验,飞蝗两型转变的关键基因已被鉴定出,但靶向调节这些关键基因的miRNA以及其分子调节机制目前尚未明确。针对这一科学问题,基于本实验室飞蝗型变的前期研究结果,我们拟综合运用分子生物学、生物化学、昆虫行为学、生物信息学和基因组学的技术,来深入探讨miRNA在表型可塑性中的调控作用。通过型变相关miRNA鉴定、体外荧光素酶miRNA靶标验证、体内功能获得/缺失性miRNA作用研究,来揭示飞蝗两型转变过程中的miRNA分子调节机制。本项目的实施将深化飞蝗型变过程miRNA调控机制的理解,为开发新的蝗灾防治策略和方法提供理论依据。

英文摘要: Understanding the density-dependent phase change between solitarious and gregarious locusts remains a hot topic in the filed of phenotypic plasticity. miRNAs are non-coding but functional RNA molecules, which regulate a diverse collection of biological processes by post-transcriptionally repress the expression of genes by base-pairing with the target mRNAs. Critical genes related to locust phase changes have been found by RNAi experiments, but the regulation mechanism of miRNAs targeting these critical genes remains unknown. To answer this question, based on the previous results of locust phase change, we will take an integrated strategy, using multidisciplinary approaches including molecular, biochemical, ethological, bioinformatic and genomic analysis. We aim to characterize the mechanism that miRNAs regulate phenotype switching of two phases of locusts, starting from identifying key miRNAs, then validating miRNA target by luciferase assay, and uncovering miRNA gene function by gain/loss-of-function experiments in vivo. Upon completion, this project will provide new insights into the miRNA-mediate regulation underlying locust phase change and facilitate to develop novel pest control methods and strategies for locust plagues.
