中文摘要: 颌骨牵引是颌面部畸形矫治的重要手段,也是机械力在生物再生领域的经典应用。间充质干细胞是骨再生细胞的重要来源,间充质干细胞在机械力刺激下可以分化为骨基质形成细胞,并分泌促进骨形成的生长因子。TRPM7是瞬时电位受体家族重要成员,是一类典型的细胞机械力感受器,我们课题组前期研究发现,机械力刺激可以活化TRPM7,介导颌骨间充质干细胞体外成骨。因此我们假设TRPM7阳性的间充质干细胞,在颌骨牵引过程中发挥机械力感受器作用,感受机械力刺激、促进牵引成骨。我们拟以单纯截骨4mm骨缺损为阴性对照;以单纯颌骨牵引为空白对照;分别在牵引组和单纯截骨缺损组注射TRPM7阳性间充质干细胞和TRPM7阴性间充质干细胞,观察间充质干细胞下游机械力相关信号通路活化情况以及牵引成骨效果。同时结合体外加力实验,进一步验证TRPM7阳性间充质干细胞能够感受牵张力刺激并促进成骨。

英文摘要: Distraction osteogenesis is widely used to reconstruct maxillofacial deformity. MSCs (mesenchymal stem cells) are the source of bone forming cells. Stimulated by mechanical stimulation, MSCs could differentiate into bone matrices forming cells and secret promoting bone formation cytokines. TRPM7 is an important member of TRP (transient potential receptor) superfamily which are widely reported to be a cellular mechanical sensor. Our previous study showed that TRPM7 in mandibular bone marrow MSCs could sense mechanical stimulation and promoting osteogenic differentiation. Thus, this project aimed to study whether TRPM7 positive mesenchymal stem cells could sense mechanical stimulation promoting their osteogenic differentiation and bone formation in mandibular distraction osteogenesis.
