中文摘要: 逆转录病毒的包装和释放一直是逆转录病毒生活周期的研究热点。近年来,发现转运必需内吞体分选复合物 (Endosomal Sorting Complex Required for Transport,ESCRT) 在逆转录病毒复制周期的包装与出芽环节起到关键作用。Citron kinase是一种GTPase RhoA的效应物,我们的研究发现citron kinase可以促进HIV-1病毒的释放,但是具体作用机制尚不清楚。在项目中计划深入探讨citron kinase促进HIV-1病毒颗粒包装释放的作用机制以及可能影响它发挥功能的其它宿主蛋白, 进一步研究citron kinase对其他包膜蛋白以及非包膜蛋白的作用从而更加深入理解病毒与宿主相互作用。

英文摘要: The assembly and release of retroviruses is the key step in retroviral life cycle. In recent years, Endosomal Sorting Complex Required for Transport (ESCRT) has been demonstrated to play an essential role in the process of retroviral assembly and budding. Citron kinase is an effector of RhoA. Our previous studies revealed that citron kinase enhanced the production of human immunodeficiency virus type-1 (HIV-1), but the mechanism remains elusive. Herein we further elucidate the detailed mechanim of citron kinase and some host proteins which probably affect its function in HIV-1 budding, and determine the effect of citron kinase on other viruses, in order to understand the interaction of host and virus thoroughly.
