中文摘要: 本项目旨在发展一种基于共轭聚合物与荧光蛋白之间荧光共振能量转移机理的高通量抗生素筛选新方法。在对共轭聚合物与不同颜色荧光蛋白(绿色、黄色、红色荧光蛋白)之间单重、多重荧光共振能量转移过程及机理系统研究基础上,以实验构建的表达不同颜色荧光蛋白的耐药菌为模型,通过分析共轭聚合物与荧光蛋白之间能量转移过程的变化来评价抗生素的敏感性,进而实现对抗生素的高通量筛选。这种筛选方法还可以拓展用于抗肿瘤药物的敏感性分析及高通量筛选。通过开展共轭聚合物与荧光蛋白之间高效能量转移过程与抗生素或抗肿瘤药物敏感性之间的相关性的研究,为深入研究细菌和细胞的耐药机理及快速筛选药物提供实验依据。

英文摘要: The object of this proposal is to develop a fluorescence resonance energy transfer (FRET) based optical approach for high-throughput antibiotics screening which employ the signal amplification properties of conjugated polymers. FRET processes and mechanism between conjugated polymers and different fluorescent proteins (green, yellow, red fluorescent protein) will be investigated firstly. With these data in hand, we carry out studies on evaluating antimicrobial susceptibility by measuring FRET ratios with drug-resistant bacteria as a model, and high-throughput screening of antibiotics will be achieved. Utilizing the similar detection principle, drug-resistant tumor cells are chosen as screening model to achieve high-throughput screening of anticancer drugs. It is believed that by monitoring the energy transfer processes of conjugated polymers and fluorescent proteins, studies on their scientific relationship with antibiotics or anti-tumor drugs screening will provide useful data for in-depth studies on the mechanism of drug resistance of bacteria and tumor cells and rapid drug screening.
