中文摘要: 生物标志物作为一种临床客观测定和评价的生化指标,有助于对生理、病理或治疗过程进行判断,是目前临床研究的基础和开发新治疗方法的着眼点。为了对生物标志物实施高灵敏和高特异地检测,已经有大量方法对其进行了研究。表面增强拉曼光谱(SERS)具有所需样品少,反映结构信息量大,高灵敏度和高分辨率的优势,能够超灵敏地检测疾病特异性的生物标志物, 为人们对于疾病的鉴定、早期诊断及预防、治疗过程中的监控起到帮助作用。申请者拟发展基于SERS检测生物标志物的技术,研究发展可以超灵敏高特异性地检测蛋白质、核糖核酸(miRNA)等生物标志物的技术,探索这种基于SERS检测生物标志物技术在复杂生物样品体系、细胞内应用,建立检测一类生物标志物的标准方法,最终将其应用于临床快速、准确、灵敏地诊断各类疾病。

英文摘要: A biomarker is defined as a characteristic that is objectively measured and evaluated as an indicator of normal biological processes, pathogenic processes, or pharmacologic response to a therapeutic intervention. At present, biomarker is a basic of clinical research and a focus of new therapeutic approaches development. Many methods for high sensitive and specific detecting biomarker had been studied. Surface-enhanced Raman spectroscopy(SERS) has advantages of low sample require, high degree of structural information about the molecule, high sensitive and selective. Biomarker detection based on SERS was beneficial for disease identification, early diagnosis and prevention, treatment process monitoring. A series of SERS detection methods with sensitive and specific will be developed for biomarker such as protein, ribonucleic acid (micro RNA). These technologies will be apply in complex biological samples and cells for standard method establishment. It is expected to be used for rapid, accurate, sensitive to the clinical diagnosis of various diseases.
