中文摘要: 干细胞移植治疗急性心肌梗死是一项非常具有积极意义的治疗手段。分子影像技术是研究移植干细胞在体内分布、评估干细胞治疗效果的重要技术手段。本研究采用带有荧光、核医学及磁共振多重标记的小鼠骨髓间充质干细胞,在功能化自组装多肽纳米纤维辅助下,移植治疗急性心梗。通过体外、体内一系列研究:评估多重标记对干细胞存活、分化的影响;结合磁共振空间分辨率高、荧光及核医学敏感性高的优势,对移植干细胞进行在体多模态示踪,研究细胞在体内的分布、存活及凋亡情况;评估自组装多肽纳米纤维支架对于提高干细胞存活的价值;同时利用磁共振及核医学成像,测定心功能及心肌代谢情况,评估干细胞及自组装多肽纳米纤维支架的治疗效果。通过本项目的研究,实现多模态在体示踪干细胞的存活及凋亡,评估干细胞在不同辅助环境下的治疗效果,为今后临床心肌梗死的细胞疗法研究提供重要参考。

英文摘要: Stem cell therapy has showed great potential in acute myocardial infarction (AMI) treatment. Molecular imaging is one of the most important techniques to study distribution of stem cells and efficacy of the treatment. In our study, we will transplant tri-labeled mice BMSCs cultured in self-assembling peptide nanofiber hydrogels into AMI mice myocardium, and monitor stem cells by optical imaging, MRI and radionuclide imaging. Firstly, through a series of study both in vitro and in vivo, we aim to evaluate the impacts of tri-labeling on stem cells survival and differentiation. Secondly, combining the high spatial resolution of MRI, and high sensitivity of optical imaging and radionuclide imaging, biological behavior of stem cells including survival, distribution and apoptosis will be monitored. Thirdly, additional benefits of self-assembling peptide nanofiber hydrogels to stem cells survival will be evaluated. Finally, cardiac function and metabolism will be measured by MRI and radionuclide imaging to assess efficacy of stem cell therapy. Our study will provide a solid base for clinical study of stem cell therapy of AMI.
