中文摘要: 人工玻璃体是治疗严重玻璃体视网膜疾病的必需品,然而具有良好生物相容性,并能长期在眼内填充的理想人工玻璃体至今仍是空白。我们前期通过体外交联获得与自然玻璃体性能相似的聚乙烯醇【PVA:poly(vinyl-alcohol)】水凝胶,并将其注入体内进行长达半年的观察发现,其拥有良好的光学物理性能、视网膜支撑功能和生物相容性。本课题拟进一步研究通过注入液态PVA水溶液和壳聚糖与β一甘油磷酸钠交联剂,在37℃条件下原位交联形成水凝胶,同时负载5-氟尿嘧啶PLGA微球,进行玻璃体相关性能检测,模拟自然玻璃体的性能;并通过兔增殖性玻璃体视网膜病变(proliferative vitreoretinopathy,PVR)模型验证其安全性和有效性,为寻找一种更为接近自然玻璃体性能的人工替代物提供实验依据

英文摘要: Artificial vitreous body is needed for the treatment of severe vitreous retinopathy, but an ideal artificial vitreous body with good biocompatibility,and can fill in the eye for a long time is still blank. Our initial study throughdiplomatic couplet body which similar to natural vitreous properties of poly (vinyl alcohol) (PVA) hydrogel, and injected into the body for half a year's study found that it has good optical properties, retina support function and biocompatibility.This topic proposed further study by injecting liquid PVA aqueous solution, and chitosan with beta glycerin sodium phosphate crosslinking agent, under the condition of 37 ℃ in situ crosslinking hydrogels formation, at the same time load 5 - fluorouracil PLGA microspheres, Detection of vitreous related parameters, restore the physiological functions of vitreous, simulate the natural function of vitreous body, And through the incremental rabbit vitreous retinopathy (proliferative vitreoretinopathy, PVR) model to verify the safety and efficacy, to find a more similar to natural vitreous properties of artificial substitutes to provide experimental basis.
