中文摘要: 口腔上皮鳞癌来源于口腔癌前病变—#8212;口腔粘膜白斑,其癌变率高达10-36% 。本项目通过比较口腔正常、白斑、癌变白斑的miRNA表达谱,发现了可能与白斑癌变相关的特异性miRNA,为进一步研发筛选白斑癌变的标记物提供了有力依据。运用QRT-PCR和免疫荧光原位杂交技术,我们验证了芯片的数据可靠性。而且利用免疫荧光原位杂交我们发现了miR-31*主要高表达在炎症、血管和肿瘤区域。此外,生物功能学研究显示,miR-31*对头颈部癌症细胞Cal-27具有抑制细胞增殖、增加细胞凋亡、抑制细胞迁移和浸润的调控功能。而对白斑细胞系Leuk-1,miR-31*仅具有显著的促凋亡作用。上述结果提示,miR-31*在口腔白斑癌变过程中具有重要作用,在不同恶性程度的口腔细胞中具有不同的调节效应。

英文摘要: Oral squamous cell carcinoma develops from oral premalignancy__oral leukoplakia which the rate of mallignant transformation is about 10-36%. In the present study, we found the leukoplakia-malignant transformation specific miRNAs by comparing the miRNA profiles of oral normal mucosa, leukoplakia, malignant transformed leukoplakia. The data provide the evidence to further develop markers of screening malignant transformed leukoplakia. We verified miRNA microarray data using QRT-PCR and FISH. Furthermore, we showed that miR-31* mainly expressed in the inflammatory and vascular area and tumor nest by FISH. Moreever, miR-31* presented the effect of inhibiting proliferation, increasing apoptosis, inhibiting migration and invasion in Cal-27 by biological analysis. On the other hand, miR-31* only showed the significant effect of increasing apoptosis in leukoplakia cell line (Leuk-1). Taken together, miR-31* play an important role in the process of leukoplakia malignant transformation. It may have different modulating effect in the cells with different malignant potential.
