中文摘要: 染色体不稳定性(CIN)与肿瘤的发生和发展密切相关,而有丝分裂检查点调控异常可促进CIN,最终促进肿瘤的发生。本室近年关注该领域,发现CUEDC2可通过促进染色体不稳定性导致肿瘤发生(NCB, 2011,共同第一作者); Cdc5L可参与纺锤体检查点的调控 (Cell death& disease ,2013,共同作者)。基于周期调控蛋白筛选得到NF-κB activating protein(NKAP)蛋白,发现其在卵巢癌中低表达,并且干涉NKAP可导致染色体错误排列,染色体分离滞后,最终促进CIN的产生。本项目在此基础上通过质谱鉴定NKAP在M期动态结合的蛋白质,利用稳定干涉、过表达细胞系以及动物模型,结合核型分析、免疫荧光等手段,进一步研究NKAP在染色体不稳定性及纺锤体检查点调控中的分子机制,并揭示其在卵巢癌发生发展中的作用及机制。这将对卵巢癌的早期诊断和治疗有着深远的意义。

英文摘要: Chromosomal instability (CIN) is closely related with initiation and development of cancer, and abnormal regulation of mitotic spindle assembly checkpoint can promote CIN and tumorigenesis. Recent years we concerned about this area, and we found overexpressed-CUEDC2 can promote CIN and tumorigenesis (Nature cell biology,2011,co-first author);Cdc5L can involve in mitotic spindle assembly checkpoint regulation (Cell Death & Disease, 2013,co author). Based on screen for cell cycle regulated protein, we found NF-κB activating protein(NKAP),which express low in ovarian cancer. Knock-down NKAP can induce chromosomes misalignment and lagging in mitosis. As we known, cells with lagging chromosome has a trend of CIN. We use mass to identify NKAP-interacted proteins specific in M-phase. Stable-expression or kown-down cell line and animal model combined with karyotyping and Immunofluorescence were also used for the research. This project will uncover the mechanism of NKAP in CIN and spindle assembly checkpoint regulation,and show the role of NKAP in ovarian cancer initiation and development. This will have far-reaching meaning on early diagnosis and treatment of ovarian cancer.
