中文摘要: 目前对PID1基因的研究主要以人和鼠为对象,对猪PID1基因的研究较少。猪PID1被发现与猪肌内脂肪沉积存在关联性,但其对猪肌内脂肪沉积的影响及其作用机制尚不清楚。基于此,本项目在克隆猪PID1基因、构建真核表达载体、制备重组猪PID1蛋白和分离鉴定猪肌内前体脂肪细胞的基础上,进一步通过动物饲养试验研究重组猪PID1蛋白主动免疫猪后对猪肌内脂肪含量的影响,并采用体外细胞培养实验研究过表达猪PID1对猪肌内前体脂肪细胞增殖和分化的影响,同时检测PPARγ、HSL、C/EBPα、FAS基因mRNA 和蛋白水平表达的变化,以探讨PID1对猪肌内脂肪沉积的影响并初步揭示其作用机理。完成本研究,不仅可为肉品质的改善提供理论依据,还可为进一步揭示猪肌内脂肪沉积的机理奠定了重要的分子基础。

英文摘要: At present, the PID1 gene studies mainly focus on humans and rats and there are few researches about porcine PID1 gene. Although porcine PID1 has been found to be correlated with porcine intramuscular fat deposition, the effects of PID1 on porcine intramuscular fat deposition and its underlying mechanisms are not clear. Based on this, in this project we will clone the porcine PID1 gene, construct the eukaryotic expression vector, prepare the recombinant porcine PID1 protein, and isolate and identificate the porcine intramuscular preadipocyte. On this basis, further in vivo studies will be performed to investigate the effects of active immunization against recombinant PID1 protein on the content of intramuscular fat, and in vitro studies will be conducted to examine the effects of porcine PID1 on porcine intramuscular preadipocyte proliferation and differentiation and the expressions of PPARγ, HSL, C/EBPα and FAS at mRNA and protein levels. The present project is of importance not only to provide theoretical evidence for improving the meat quality, but also to establish important molecular basis for revealing the mechanisms of intramuscular fat deposition.
