中文摘要: 肾间质纤维化是慢性肾脏病进展的显著特征,肾小管基底膜完整性的破坏在肾脏纤维化过程中发挥关键作用。组织纤溶酶原激活剂(tPA)/基质金属蛋白酶9(MMP9)是调节基底膜完整性的重要因素。促红细胞生成素(EPO)能延缓肾脏纤维化,但其机制不明。有研究发现,EPO能上调血浆miR-144的水平,而血浆中的miRNA大都存在于微囊泡中,miR-144的靶基因是tPA。本项目以单侧输尿管梗阻(UUO)的肾组织和肾间质成纤维细胞为研究对象,观察EPO注射后血浆中包含miR-144的微囊泡含量的变化,以及包含miR-144的微囊泡对活化的成纤维细胞中tPA/MMP9的表达和活性的影响,并研究miR-144对tPA的调控机制,探讨EPO是否通过上调血浆中包含miR-144的微囊泡,抑制成纤维细胞tPA/MMP9的表达和活性,进而保护小管基底膜的完整性,延缓肾组织纤维化。

英文摘要: Chronic kidney disease is characterized by tubulointerstitial fibrosis. Disruption of the integrity of tubular basement membrane (TBM) contributes to renal fibrosis. Erythropoietin (EPO) attenuates renal fibrosis; however, the mechanism remains unknown. It was reported that serum miR-144 was up-regulated after EPO. Most of the serum miRNAs are found in microvesicle. tPA is the target of miR-144. Here, in UUO model and cultured fibroblast, miR-144 level in serum microvesicles will be evaluated after EPO administration. The effect of miR-144-containing microvesicle on tPA/MMP9 expression and activity will be investigated. Further, regulation mechanism of miR-144 on tPA expression will be determined. The aim of this study is to investigate whether EPO protects the integrity of TBM by up-regulating serum level of miR-144-containing microvesicles, which then inhibit the expression and activity of tPA/MMP9 in fibroblast.
