中文摘要: 鼻咽癌作为华南及东南亚地区的高发恶性肿瘤,其远处转移是鼻咽癌治疗失败的主要原因。我们前期研究证明IGFBP-6能够抑制鼻咽癌细胞株的生长增殖,体内动物实验显示IGFBP-6基因干扰后能够促进鼻咽癌细胞的远处器官的转移,其表达是晚期鼻咽癌患者远处转移的相关因素。有研究表明IGFBP-6能够与IGF-Ⅱ紧密结合,发挥对肿瘤的生物学调节作用。但IGFBP-6/ IGF-Ⅱ调控鼻咽癌转移的机制未明。我们希望通过IGFBP-6稳定转染低表达IGFBP-6的鼻咽癌低转移株6-10B,从信号转导通路方面探讨IGFBP-6调节IGF-Ⅱ的分子机制;细胞运动、侵袭等实验及体外实验模型观察IGFBP-6/ IGF-Ⅱ调控鼻咽癌转移的作用,检测鼻咽癌组织及血清中IGFBP-6及IGF-Ⅱ的表达的相关性及对远处转移的影响。为阐明鼻咽癌转移的分子机制奠定一定的理论基础,并为寻找新的候选靶分子提供一定的理论基础。

英文摘要: Nasopharyngeal Carcinoma (NPC) is endemic in Southern east Asia and Southern China. The main cause of therapy failing is distant metastasis. We have found that IGFBP-6 could inhibit the proliferation of NPC cells and the IGFBP-6 gene interference could increase the ability of distant metastasis of NPC cell in vivo. The expression of IGFBP-6 was an related factor for NPC patients metastasis. Some study have showed that IGFBP-6 can closely integrate with IGF-Ⅱ and cause the regulating biological function. But the mechanism of IGFBP-6/ IGF-Ⅱregulate NPC is unclear. After the stable transfer IGFBP-6 in the NPC cell line 6-10B, we hope to study the molecular mechanism of IGFBP-6 regulate IGF-Ⅱ, the cell moving and invasion from the in vivo animal model. We also hope to detect the relationship between the IGFBP-6 and IGF-Ⅱ effected by the distant metastasis. Our study will make the molecular mechanism clear about NPC metastasis and provide the base for the new candidate target molecules.
