中文摘要: 细胞衰老是一个细胞周期抑制和细胞生长停滞的生理过程。细胞衰老是细胞抑制肿瘤发生发展的主要机制之一。在肿瘤和衰老研究领域中,学术界同时存在线粒体假说和端粒假说,但对这两种假说之间的联系却知之甚少。我们前期的研究也表明TIN2作为一个关键的端粒体蛋白,不仅在端粒维持上起关键作用,也参与调节线粒体中的呼吸作用及细胞代谢。我们将系统研究TIN2及其相互作用蛋白在端粒维持、细胞代谢、细胞衰老信号传导和肿瘤发生发展等重要生物学过程中的作用。开展TIN2及其蛋白调控网络的研究不仅有助于我们揭示端粒维持和细胞能量代谢之间关系的理解,而且对我们全面了解细胞衰老和肿瘤发生的机制,开发以端粒相关蛋白为靶标的抗肿瘤药物具有重要意义。

英文摘要: Cellular senescence is a physiological process of cell cycle arrest and cell growth stagnation, and represents one of the main mechanisms to inhibit tumorigenesis and cancer development. In the field of cellular senescence and tumorigenesis, there are several major hypotheses including the mitochondrial hypothesis and telomere hypothesis. However, the relationship between these two hypotheses remains poorly understood. Our previous studies have shown that telosome/shelterin protein TIN2, not only plays an important role in telomere maintenance, but also involves in the regulation of mitochondrial respiration and cellular energy metabolism. We will systematically study the function of TIN2 and its interacting proteins in telomere maintenance, cellular metabolism, cell senescence signaling, and tumor initiation and development. Studies of TIN2 and its regulatory network will reveal the relationship between telomere maintenance and cellular energy metabolism, aid our undersatnding of molecular mechanisms of cell senescence and tumorigenesis,and help to develop anti-tumor drugs that target telomere related protein.
