中文摘要: 多个miRNA(微小RNA)在癌症中协同工作,调节癌变,对于肿瘤的发生发展有重要作用。本课题以miRNA网络为研究对象,以发现在癌变中协同工作的miRNA为研究目标,探索miRNA调控方向确定的方法、多miRNA协同工作的机理及预测未知协同的技术,从而了解癌变中基因的变化规律和相互影响,进而指导治疗和药物设计。首先,基于信息熵发现miRNA网络中两个miRNA之间的调控方向;其次,基于自组织理论,扩展某一癌症中的miRNA关联对到任两个miRNA,发现它们之间的最短协同路径,构成协同机制候选集;再次,基于高维核密度,评估候选集,发现重要的协同机制;最后,基于模糊集和统计学,在信息不完全环境下结合癌症种类预测未知机制与癌症的相关信息。本课题预期在概念类关联、自组织寻径、高维核密度和不完全信息聚类等方面有理论和算法上的创新;同时对癌变病理分析提供基因层面的技术支持。

英文摘要: Cancer inevitably involves miRNAs (microRNAs) for multiple miRNAs can regulate the development of cancer by cooperating each other as a directed synergistic regulation network. However, how to identify the regulation directions of miRNAs and master the principles of miRNA synergistic mechanisms is still remaining. So, this project concentrates on the directed synergistic regulation mechanisms of miRNAs in cancer based on miRNA network. We aim for the determination of miRNA regulation directions, the principles of multi-miRNA synergisms, as well as the prediction of unknown synergisms. Our research strategy is detailed as follows. According to this goal, our plan first constructs an miRNA network based on sequence and secondary structure of miRNAs. Then, regulation direction of two miRNAs will be identified by entropy theory. Next, such directed relations will be extended from two miRNAs to more, during which self-organized technology is employed. This step can discover the directed shortest pathway of any miRNA pairs, composing the candidate synergism set. Further more, we evaluate this set using high-dimensional kernel density estimate, in which the weak relations of miNRAs will be filtered so as to verify the mature directed synergistic regulation mechanisms.
