中文摘要: Innexin (Inx)蛋白是一个重要的昆虫免疫蛋白。前期的研究已经表明Inx在不同的细胞中分别有促进细胞凋亡和聚集的功能,与pI3K/Akt磷酸化相关。本项目抓住Inx蛋白以Inx单体,Inx6聚体(Hemichannel/Innexon)和Gap junction (GJ)存在细胞中执行三种功能,这一关键问题,提出科学假说:Inx参与到昆虫细胞免疫的结节、凋亡和包囊反应中。本申请拟采用转录组学、分子生物学和昆虫免疫学的方法和手段加以实验验证, 即Inx的表达促进pI3K/Akt去磷酸化,定位于细胞核的Inx与周期蛋白CCND相互作用,调控细胞聚集;位于细胞膜表面的Innexon,激活caspase3,导致细胞凋亡;位于相邻两个细胞表面的GJ,传递上述两种细胞信息,在包囊形成过程中产生细胞的聚集和凋亡。这个假说的证明将促进昆虫细胞免疫的研究,为害虫的生物防治提供新的免疫分子靶标。

英文摘要: Gap junction (GJ), specialised membrane structures that mediated intracellular communication through forming gap junction channel between two neighboring cells, intracellular and extracellular communication by forming hemichannel (innexon) in the cell surface is well known in the vertebrate. A few studies also report that GJ also regulate cell cycle in the cell nuclei by single protein. GJ terms innexin (Inx) in the invertebrate. Previous study shows that inx promote cell apoptosis and aggregation in different insect cell type involving in the dephosphorylation of PI3K/Akt signaling pathway. Based on previously studies and most recent reports, we propose a hypothesis that Inx may perform a triple function to regulate nodule and aggregation during insect innate cellular immunoresponse, also regulate cell apoptosis. To test hypothesis, we will employ transcriptome sequencing, molecular biology technology and combine insect immunity methods to test this hypothesis: single Inx localizes in the cell nucleus to regulate aggregation via cyclin D (CCND); Innexon localizes in the cell surface to regulate cell apoptosis via dephosphorylation of pI3K/Akt to activate caspase 3. GJ is responsible for transmitting two information, aggregation and apoptosis, between two cells during encapsulation forming. This study will expand understanding of innate cellular immunoresponse. In addition, the results will be a good source for screening of novel molecular target gene for biology control.
