中文摘要: 基于液滴的微流控芯片使用不相溶的两相流体在微孔道界面处形成微小液滴,并以液滴为载体进行生化反应或检测。液滴微流控系统由于具有体积小、低扩散、无交叉感染、反应快速等特点近年来迅速成为研究热点。本项目首次将液态金属液滴引入微流控芯片中开辟一个新的液滴微流控研究方向。由于液态金属导电率高,热导率高,比热小,蒸发慢,与大多数流体不相溶,非常适合作为生化反应特别是温度敏感类型反应的载体(例如PCR)。本项目将探索液态金属液滴在微流控芯片中传热传质的机理,建立一个集液态金属液滴的生成、样品装载、液滴融合、温度控制、液滴检测、液滴收集等功能于一体的微流控芯片系统。此外还会建立一个基于开放性液态金属液滴作为载体的超快速PCR系统。本研究属于世界领先的微流控概念,基于液态金属液滴的微流控系统的概念在国际上还尚无人提出,此系统有望大大提高PCR速度。其实施兼具十分重要的理论学术意义和实际应用价值。

英文摘要: Droplet-based microfluidic chip is a hot spot for researches in recent years. Two or more immiscible fluids meet inside a droplet-based microfluidic chip, and droplets are generated at the interface, always from nanoliter to picolitter in volume.Droplet-based microfluidic chip has many merits, such as small volume, low dissipation, no contamination, high reaction speed and etc. This project will indroduce liquid-metal droplet into the droplet-based microfluidic system. Liquid metal (such as gallium or eutactic of gallium and indium) has high electric/heat conductivity, low heat capacity, low evaperation and doesn't mix with almost all the other normal liquids, which is very suitable for on-chip temperature dependant reactions. This project will explore the mechanism of heat and mass transfer of liquid-metal droplet from microscale point of view.A system integrated with droplet generation, sample loading, droplet combination, temperature control, droplet detection and droplet sorting will also be built. Finally, an open liquid-metal droplet based ultrafast PCR system will also be developed. This project proposed a new concept of liquid-metal droplet based microfluidic system that can lead currenty related research in the world.
