中文摘要: 对深层生物组织细胞内微环境进行准确动态表征是生物学和生物医学等领域的重要目标之一。由于活体生物样品表面的不平整性和组织内部折射率分布的不均匀性等原因,使得成像系统像差较大,获得的图像质量较差,且随着成像深度的增加像差越严重。在本项目中,我们结合双光子激发荧光显微成像技术、AOD扫描技术、FLIM技术和COAT像差校正技术,发展一种针对深层组织细胞的动态荧光寿命成像平台,用于实现对深层组织细胞内蛋白质等相关参数的定量测量和动态监测。

英文摘要: The accurate and dynamic characterization of deep biological tissue microenvironment is one important goals in the fields of biology and biomedical. Because the surface irregularities 、the within uneven distribution of the refractive index and other reasons for living organisms sample, so that the imaging system has lager aberrations, and get poor image quality. And with the increasing of the imaging depth, the aberration become more serious. In this project, we combine two-photon excitation fluorescence microscopy imaging technology、AOD scanning technology、FLIM technique and COAT aberration correction technology to develop a platform of dynamic fluorescence lifetime imaging for deep-tissue cells, and use the imaging system to implement the quantitative measurement and dynamic monitoring of proteins and so on related parameters in deep tissue cells.
