中文摘要: Antisense RNA对基因表达调控具有重要作用。第二代链特异转录组测序技术促进了非编码RNA研究。申请人前期基于小鼠多组织细胞链特异性RNA-seq数据和表观修饰数据,进行了非编码RNA鉴定及表观修饰研究。本项目拟在前期研究基础上,增加小鼠肝和心脏等组织RNA-seq数据,整合公共数据,通过多组织和细胞RNA-seq数据分析,构建小鼠基因表达谱;鉴定Cis-和Trans-antisense RNA;研究Antisense RNA的特征及其与表观修饰的关系;揭示正义、反义链RNA表达相关性;分析Antisense RNA靶基因功能;构建小鼠多组织和细胞Antisense RNA数据库并提供在线Antisense RNA鉴定及分析。本研究对于注释小鼠非编码RNA(特别是Antisense RNA)、分析Antisense RNA特性、揭示正义、反义链RNA表达调控关系等具有重要意义。

英文摘要: The recent studies suggest that antisense transcripts play the important regulatory role in gene expression and the next generation sequencing technologies promote the study of non-coding RNA. In our lab, the applicant did non-coding RNA identification and epigenetic modification study based on the strand-specific RNA-seq data and epigenetic modification data for mouse. In this study, based on the preliminary studies, we will do RNA-seq for liver, heart and other important tissues, integrate public, old and new data, and do analysis for them as following: obtaining the gene expression profiles; identification cis- and trans-antisense RNA; study the characteristics of antisense RNA and the relationship between antisense RNA and epigenetic modifications; finding the association between sense and antisense RNA; analysis the function for target genes of antisense RNA; building antisense RNA database using these data, and providing antisense RNA identification and analysis for RNA-seq data based on web service. This project is very important and significant for annotation the non-coding RNA (especially for antisense RNA), anaylsis the characteristics of antisense RNA, and finding the regulating relationship between sense and antisense RNA in mouse.
