中文摘要: CD82被认为是广谱的肿瘤转移抑制因子。对其作用机制的研究日益受到重视。但其详细分子机理仍不清楚。其转移抑制作用分子结构基础的研究,对阐明CD82抑制肿瘤转移的分子机理,开发抗肿瘤转移新药,具有十分重要的理论意义和潜在应用前景。我们前期对CD82胞外结构域功能研究中,发现CD82胞外小环结构域多肽(CD82-SEL)对多种组织来源的肿瘤细胞,包括大肠癌细胞、乳腺癌细胞和肝癌细胞的运动迁移能力均有强烈的抑制作用,在50μg/ml 浓度时,细胞体外迁移完全被抑制。此属首次报导。其作用机理以及在体内对肿瘤转移抑制作用尚不清楚。该课题将对CD82-SEL多肽抑制癌细胞运动迁移的详细机理进行研究。观察CD82-SEL多肽在动物体内对肿瘤细胞转移的抑制作用。并通过对CD82-SEL多肽N-端、C-端脂酰化修饰,研究其对肿瘤细胞转移作用的影响。为将CD82-SEL开发抗肿瘤转移新药提供数据。

英文摘要: CD82 is considered as a wide-spectrum invasion- and metastasis-suppressor.The study on the mechanism of CD82 inhibiting cancer metastasis has been paid more and more attention.But the precise mechanisms are still unclear.The study on the molecullar structure foundation of CD82 will play an important role in the ilucidating the mechanism of CD82 inhibiting cancer metastasis and developing new anti-metastasis drugs.In our previous studies on the mechanism of CD82 inhibiting cancer metastasis, we found that CD82 small extracellular loop domain can markedly suppress the in vitro migration of cancer cells from different tissue ,including colorectal,breast and liver, 50μg/ml of CD82-LEL polypeptide can completely inhibit the cell migration. In this project, we shall further investigate the mechanism of CD82-SEL inhibiting cell migration, test the role of CD82-SEL anti-metastasis in vivo and or esterificate the CD82-SEL peptide at N-and C-terminal.The study of project will present the valuable date for developing anti-metastasis drugs from CD82-SEL peptide.
