中文摘要: 恶性肿瘤严重威胁着人类的健康,而肿瘤标志物直接反映肿瘤是否存在,实施肿瘤标志物高灵敏的早期检测对于肿瘤的诊断和预防极其重要。 灵敏度是电化学肿瘤标志物检测的关键因素。本项目拟将正电性的纳米金种子引入双螺旋DNA负电性的磷酸骨架中形成长度可控且均匀的新型金纳米线桥,并应用于肿瘤标志物的电化学检测,实现检测信号的多重放大。同时,精心设计两条交错性互补的DNA序列,利用DNA碱基互补原理促使DNA杂交链式反应发生,实现DNA模板的n倍循环增长,通过引入更多金种子建立类似于PCR放大的高灵敏电化学肿瘤标志物检测新方法。此外,将不同的DNA序列分别固定在信号探针标记的不同肿瘤标志物对应抗体上,实现多蛋白血清混合体系中目标蛋白的高选择性检测和多种蛋白的同时检测,拓展其临床的应用价值。

英文摘要: Malignant tumors have serious threat to human health, and tumor markers directly indicate the presence of tumors, constructing highly sensitive detection of tumor markers are of great importance in the diagnosis and prevention of tumors. Sensitivity is the key point of tumor markers electrochemical detection. This project aims at introducing positively charged gold seeds to negatively charged phosphate backbone of double strand DNA for the formation of length-controled and uniform gold nanowire bridge, and using them to construct tumor markers electrochemical detection, realize the multi-amplification of electrochemical signal. Meantime, designing two strands of interleaved complement DNA to induce DNA hybridization chain reaction (HCR) happen according to watson-crick base pairing, realize the n times cycle amplifying of DNA template, highly sensitive PCR-similar tumor markers electrochemical detection method is prepared based on introducing much more gold seeds. In addition, different DNA sequences are designed and immobilized onto the antibody suface of different tumor markers to construct highly selective detection of target protein in mixed serum solution and the simultaneous determination of two proteins, broden the application of them in clinical assay.
