中文摘要: 在近些年的分析科学研究中,功能核酸已经成为构建生物及化学传感器的一种重要元件。相应的功能核酸传感器由于信号报导方式及灵敏度的需要,往往需要引入复杂的标记步骤,增加了相应技术的应用难度。这里提出利用核酸本身具有的特性,结合脱氧核酶具有的催化功能及核酸作为荧光纳米簇生成模板的性质,在不添加标记的情况下即可获得传感信号,从而建立无标记的功能核酸传感体系。另外在识别过程中引入基于核酸酶切的信号放大机制,通过核酸酶对底物序列的水解作用释放目标物,使得目标物在该检测流程中反复作用,产生多重的信号报导,从而获得高放大倍数的信号增敏。基于以上方法并结合化学发光、比色、荧光等手段,实现简便快速的检测。该研究将有助于简化功能核酸传感技术,并扩展其应用范围,推进其的进一步分析应用。

英文摘要: In recent years functional nucleic acid has become one of the most important element in the development of biological and chemical sensors. Due to the requirements from signaling methods and sensitivity, usually tideious labeling steps are included, which increase the difficulties in the application of related technique. Here, it is proposed that some special aspects of functional nucleic acids, as the catalysis from DNAzyme and function as the template for noble metal nanoclusters, will be applied in the sensing steps and signals can be directly obtained without further labelings. Furtherly nuclease-based amplication is to be included in the sensing that muclease cleavages the strands after recognition and recycles the targets. Simple and efficinet chemiluminescence, colorimentry and fluorescence detections will be developed with the proposed mehtod. This reseach will ease the sensings with functional nucleic acids and accelertate the application of relared techniques.
