中文摘要: 本项目结合生物实验和数理建模,通过对电离辐射前后肿瘤细胞在四维时空(三维空间加时间)中生长模式的研究,为探索临床"带瘤生存"新思路做前期基础研究。有别于常规放射生物学实验,我们用体外三维细胞培养来模拟体内肿瘤细胞生长的组织力学环境,并用共聚焦显微镜跟踪拍摄细胞运动过程。我们应用软物质的实验分析技术,识别细胞空间分布,跟踪细胞运动变化,计算细胞之间相互作用。更进一步,我们运用数理手段对实验中的肿瘤细胞生长进行定量分析,并将实验分析结果综合到动力学模型中,对肿瘤细胞生长模式进行系统研究。

英文摘要: This project combined biological experiments and mathematical modeling, through the study on tumor cell growth patterns in the four dimensions (three-dimensional space plus time) before and after radiation, to make early basic research for the clinic new idea of "survival with tumors". Different from conventional radiobiological experiments, we used three-dimensional cell cultures in vitro to imitate the mechanical environment of the tissue in vivo where tumor grows, and tracked the cell movement by confocal microscopy. We applied experimental techniques of soft matter to identify the cell spatial distribution, track the cell movement, and calculate the interaction between the cells. Furthermore, we analyzed the experiments of tumor cell growth quantitatively by mathematical means, and then applied the analyazing results into the dynamic model for the systematic research on the growth pattern of tumor cells.
