中文摘要: 细胞氧化还原状态(redox status)与肿瘤、炎症、神经退行性疾病、糖尿病、心脑血管疾病等重大疾病有着重要关系。实现生物活体氧化还原状态的可视化,检测生物氧化还原状态的实时变化,对于疾病病理机制研究及开发新的诊断治疗方法具有至关重要的作用。本项目成功合成了对氧化还原环境敏感的荧光分子探针,可以在荧光、核磁共振(MRI)、电子自旋共振(ESR)等模式下检测。我们在细胞水平和活体水平上对探针的应用进行了探索。细胞实验结果显示,探针的部分性能优于商业化试剂DCFH-DA。我们应用活体荧光成像方法对皮肤氧化损伤模型进行了活体成像研究,本探针可以显示活体皮肤的氧化还原水平变化,有望在皮肤氧化损伤、皮肤癌、炎症等疾病研究中得到应用。

英文摘要: Cellular redox status is related to many diseases, such as cancer, inflammation, neurodegenerative diseases, diabetes, cardiovascular and cerebrovascular diseases. Visualization of redox status and real-time detection of redox status changes in vivo will play very important roles in the pathologic studies of these diseases. And will also be quite valuable to the development of new diagnostic and treatment methods. We have successfully synthesized some redox-sensitive probes which can be detected by fluorescence imaging, magnetic resonance imaging (MRI) and electron spin resonance (ESR).We tested and verified the probes both in cell models and animal skin models.The results showed that the probes are useful tools for studying redox status of living cells,and they can be used to study the redox changes in the skin. They are promising imaging probes for the study of skin cancer and inflammation.
