中文摘要: 糖尿病可致阴茎海绵体组织纤维化、平滑肌、内皮细胞含量减少,这与糖尿病性勃起功能障碍(ED)的发生相关。近年来国内外报道,TGFβ1-Smad信号通路过度激活与上述病理改变关系密切。我们前期研究发现,降低糖尿病大鼠阴茎海绵体内过表达的TGFβ1可减少阴茎海绵体平滑肌和内皮细胞凋亡,据此我们推测调低TGFβ1凋亡信号通路关键蛋白可能是防治糖尿病性ED的有效手段,与干细胞治疗相结合,可增强干细胞及相应功能细胞的抗凋亡能力,提高干细胞治疗糖尿病ED的疗效。为证明这种推测,本实验采用分子生物学、电生理学等技术,分细胞和整体两个层次,首先研究hHGF干预外源性TGFβ1诱导的上述细胞凋亡作用,再观察过表达hHGF的脂肪干细胞对糖尿病ED大鼠阴茎组织细胞凋亡、弹力纤维系统的影响及可能具有的神经营养作用,为糖尿病ED治疗探索新靶点,此项目国内外未见报道。

英文摘要: Diabetes may cause fibrosis and decrease in the content of smooth muscle and endothelium in corpous cavernosum, which may be correlated with diabetic erectile dysfunction(ED). Recent reports show that the over-activated TGF β1-Smad signaling pathway is closely related with above pathological changes in home and abroad. Our prior study found that down-regulation of over-expressed TGF β1 may decrease the apoptosis in smooth muscle and endothlial cell in corpous cavernosum. So we infer that down-regulation of key proteins in TGF β1 induced apoptotic signaling pathway may be effective to prevent and cure diabetic ED, which may be united with stem cell treatment to strenghten the anti-apoptotic ability of stem cell and corresponding functional cells so as to enhance the therapeutic effect on diabetic ED. In order to prove this inference, we will utilise molecular biological and electrophysiological technics on the levels of cell and animal to study the effect of hHGF on interfering the apoptosis of above cells induced by exogenous TGFβ1 firstly,then we will observe the effect of hHGF over-expressed adipose derived stell cells on apoptosis of penile cells, elastic fiber system and poassible neurotrophic effect in diabetic ED rats.
