中文摘要: 以黄精凝集素诱导肿瘤细胞凋亡与自噬分子机理研究积累为基础,基于前期构建的凝集素诱导肿瘤细胞凋亡与自噬的信号通路蛋白质相互作用网络中部分关键蛋白群为核心,集成芯片分析和生物信息学技术分析与该网络关键蛋白相互作用的miRNA, 筛选与黄精凝集素抑制肿瘤细胞增殖相关的miRNA, 结合实验进行进一步验证,获得相关miRNA的详细信息。在细胞程序性死亡相关信号通路蛋白质相互作用网络调控基础上整合miRNA对关键蛋白的调控作用,构建"凝集素-肿瘤细胞程序性死亡相关蛋白群-miRNA相互调控网络",丰富信号通路调控网络的层次,从蛋白质相互作用扩展至miRNA-蛋白质以及蛋白质间相互作用有机构成的信号通路调控网络,为最终阐述凝集素作用的精细分子机制研究提供研究模式和参考。

英文摘要: Based upon the molecular mechanisms of Polygonatum cyrtonema lectin (PCL)-induced apoptosis and autophagy in cancer cells, we will target the hub proteins identified from the PCL-modulated apoptotic and autophagic signaling protein interaction networks in cancer, take advantage of microarray and bioinformatic techniques to analyze microRNAs (miRNAs) which interact with the hub proteins in the above networks and further screen for miRNAs involved in the tumor growth inhibition effects of PCL, then, experimental verification will be performed to get more detailed information about the related miRNAs. On the basis of programmed cell death (PCD) signaling protein interaction networks, we will integrate the regulatory effects of miRNAs on hub proteins and construct the "Lectin-Hub proteins of PCD in cancer-MiRNA interaction networks", which may undoubtedly improve the structures of signaling networks and extend protein-protein interaction networks to more well-organized miRNA-protein and protein-protein interaction networks, and thus providing paradigm and reference for further elucidating the elaborate molecular mechanisms of anti-tumor properties of lectins.
