中文摘要: 丝状真菌具有卓越的蛋白分泌能力,在工业酶的生产中具有重要地位。研究丝状真菌蛋白分泌途径相关基因及调控因子的功能,有助于解析丝状真菌的高效蛋白分泌机制和进一步通过遗传改造提高菌株的蛋白分泌能力,具有重要的理论和实践意义。 本研究以工业纤维素酶生产真菌斜卧青霉为材料,对菌株的分泌压力响应和参与未折叠蛋白响应(UPR)中转录调控蛋白Hac1的作用机制开展深入研究。研究拟利用全基因组表达谱分析技术对菌株应对不同分泌压力(自身蛋白分泌压力、异源蛋白分泌压力)时的基因转录差异情况进行分析,同时结合基因靶向操作技术和染色质免疫共沉淀测序技术对Hac1的靶基因进行鉴定。通过对差异转录基因及Hac1靶基因的生物学功能进行分析,预期将对丝状真菌中Hac1的调控机制作出系统阐述,对丝状真菌应对不同分泌压力响应的分子机制获得更为深入的理解。

英文摘要: Filamentous fungi are extraordinary in their capability of secreting large amounts of proteins into the growth medium. This property has been widely exploited for producting enzymes used in industries. Functional analyses of the genes and transcription factors involved in secretion pathway will give a more detailed insight into the efficient secretion mechanism of filamentous fungi, and will provide targets for increasing their capacity of protein secretion by genetic engineering. This study will focus on the effects of secretion stress and the function of transcription factor Hac1 of unfolded protein response pathway in Penicillium decumbens. In this study, whole-transcriptome profiling and ChIp-sequencing analysis will be used to uncover the target genes regulated by Hac1. The molecular functions of the genes and the biological processes they are involved in will be investigated to get a thorough understanding of the function of Hac1. Also, whole-transcriptome analysis will be used for the determination of the differences in gene expression when the fungus is under different protein secretion stresses (from high-level native protein production or heterologous protein production).
