中文摘要: 茉莉素在植物病虫害防御和发育过程中都起着重要的作用。研究发现,茉莉素在不同植物生殖发育过程中具有特化的生物学功能;我们最近的研究结果显示,茉莉素可以通过OsJAZ1-OsMYC2-OsMADS1信号通路促进花器官起始和分生组织发育,茉莉素合成酶突变体eg1-3和茉莉素信号通路抑制蛋白突变体eg2-1D(又叫osjaz1)花器官发育过程异常,产生稃片结构、分生组织转换延迟。为深入研究茉莉素信号在水稻花器官发育的调控网络,本项目拟以eg2突变体为研究材料,利用Y2H和RNA-Seq数据,开展OsJAZ1分子调控网络的研究,通过CRISP基因定向突变、表达模式分析、遗传学和分子生物学分析,深入解析OsJAZ1调控水稻花器官发育的分子机制;另一方面,利用CRISP基因定向突变技术,分析茉莉素与其他植物激素在花器官发育过程中的互作关系,深入解析茉莉素调控水稻花器官发育的分子网络。

英文摘要: Jasmonates (JAs) are a family of compounds derived from jasmonic acid with various biological activities, which plays important role in the defense against insects and pathogens, as well as in plant development.Rrecently, jasmonic acid was revealed to play diverse roles in controlling plant reproductive development. In rice, JA signal induces the expression of OsJAZ1, a JA signalling repressor that interacts with a putative JA receptor, OsCOI1b, to trigger OsJAZ1’s degradation during spikelet development. OsJAZ1 also interacts with OsMYC2, a transcription factor in the JA signalling pathway, and represses OsMYC2’s role in activating OsMADS1, an E-class gene crucial to the spikelet development. The mutations of JA biosynthesis gene EG1, and JA signalling patway gene EG2/OsJAZ1, exhibit altered spikelet morphology with changed floral organ identity and number, as well as defective floral meristem determinacy. To futher uncover the molecular mechansim of JA in controlling rice spikelet development, we will perform yeast two hybrid (Y2H) and RNA-Seq analysis on eg2-1D/osjaz1 mutant. By using CRISP assay, we plan to investigate the regulatory network of OsJAZ1.
