中文摘要: 本课题组在前期工作中发现粒细胞集落刺激因子(G-CSF)对血红蛋白激活原代小胶质细胞中NF-κB蛋白的表达和转录活性及大鼠脑出血炎症反应有明显的抑制作用,因此认为G-CSF可能干预了脑出血中NF-κB信号转导途经介导的炎症反应,然而其确切机理尚不清楚。项目内容拟对G-CSF干预脑出血中炎症反应的作用机制进行研究。采用体内外相结合实验,通过免疫共沉淀,免疫蛋白印迹、电泳泳动度移动和流式细胞等现代分析技术,以NF-κB为切入点,探索G-CSF对NF-κB信号转导通路的调控机制,并利用自体血注入法建立大鼠脑出血模型探讨G-CSF对脑出血后小胶质细胞活性的调节机制以及对血肿的清除作用的影响。通过本项目研究可以阐明G-CSF干预NF-κB信号转导通路介导的激活小胶质细胞致炎作用的分子机制,研究结果将为G-CSF治疗脑出血提供一个新的思路和方向。

英文摘要: In previous work we found that Granulocyte Colony-Stimulating Factor (G-CSF) not only inhibits the NF-κB protein expression and transcriptional activity in microglial cells activated by hemoglobin but also suppressed inflammatory reaction and neuronal apoptosis in rats with intracerebral hemorrhage, suggesting that G-CSF may plays an important role in NF-κB mediate inflammatory responses in intracerebral hemorrhage, but the exact mechanism of is unclear. In this study we investigate the effect of G-CSF on NF-κB activation in hemoglobin-stimulated primary rats microglia, autologous blood-induced rat intracerebral hemorrhage. In addition, using immunoprecipitation, western blot, electrophoretic mobility shift assay, and flow cytometer to determine how G-CSF regulates the NF-κB signaling pathway. Furthermore, using rats intracerebral hemorrhage models to explore the role of G-CSF in NF-κB signaling pathway. This research may provide a new molecular target for therapeutic intervention in human intracerebral hemorrhage diseases.
