中文摘要: 棉纤维细胞起始数量直接决定单个棉籽上的纤维数目和棉花的产量,深入研究棉纤维起始过程关键基因,不仅对阐明纤维形成的分子机理有重要的理论意义,并可为棉花纤维的基因工程改良奠定基础。在前期研究中,我们已将陆地棉(Gossypium hirsutum)N1基因定位在At12染色体(Chr.12)体上1cM区域内,本项目拟在此基础上,采用锚定N1基因两端的分子标记,利用亚洲棉的精细基因组图谱开发新的分子标记,应用大遗传分离群体对N1基因进行精细定位,克隆并得到N1基因的全长cDNA和启动子序列。并通过基因转化、表达谱分析、启动子分析、和亚细胞定位等手段深入研究N1基因的功能,探索N1基因控制短绒起始和影响长纤维发育的分子机制。

英文摘要: Fiber initiation of Cotton (Gossypium hirsutum)has a significant impact on the number of fibers, which are major factors determining lint yield. Cloning of the key genes during fiber initiation is of great theoretical significance for elucidating the molecular mechanism of the fiber development, and lay the foundations for cotton fiber improving by genetic engineering.In our previous study, N1 gene of G. hirsutum had been mapped on At12 chromosome (Chr. 12) with a 1cM interval. Based on these results, this project will take advantage of the fine map of G. arboreum genome for developing new molecular markers involved in the genome region in which N1 gene located, and then map the N1 gene by application of a big genetic segregation population. The sequence of full-length cDNA and promoter region are obtained according to the results of map-based cloning N1 gene.Further, the function of N1 gene will be investigated by gene transformation, expression profile, promoter analysis,and subcellular localization, which would discover the molecular mechanism that N1 gene controls the fuzz-initiation and influences the fiber development.
