中文摘要: 本课题组前期研究证实,重要的环境污染源TCDD与腭部发育障碍有明显的相关性,但其作用机制尚不明确。本课题拟在前期工作基础上,以TCDD诱导胎鼠腭部发育障碍,通过体内、体外实验,观测胎鼠腭突组织和腭突细胞基因组甲基化状态,以及控制腭突融合的重要因子TGFβ3的基因甲基化程度与表达水平,并以甲基基团供体叶酸干预,以探明腭部发育障碍过程中DNA甲基化修饰的作用和机制,为预防腭裂的发生提供实验依据。

英文摘要: Our preliminary study had confirmed the correlation that the important environmental pollution TCDD relate to palate developmental disorders, but its mechanism is not clear.Base on previous work,through in vitro and in vivo experiments,the subject will establish TCDD-induced fetal mice palate developmental disorders model,and supply methyl donor folic acid,to observate the fetal mice palatal tissue and palatal cells genome methylation status,and the important palate fusion factor TGFβ3 gene methylation degree and expression level.To prove the role and mechanism of DNA methylation in the process of palate developmental disorders and provide the experimental basis to prevent the occurrence of cleft palate.
