中文摘要: 酶蛋白质是生命中必需和通用的大分子,对其结构功能的研究与新药的开发、农作物的收成以及生化合成等有着重要的联系。用实验的方法来研究酶蛋白质结构和功能本身就是低通量的,已经不能用来注释在高速发展的基因组测序技术中所获得的大量蛋白质。因此人们通过计算机技术,采用数学方法和物理模型,利用各种高通量的实验数据来进行酶蛋白质结构功能研究和预测成为必然。酶蛋白质分子的功能往往体现在关键的结合位点上,因而酶蛋白质的结构及功能位点预测也是酶蛋白质研究的热点之一。本课题主要的研究内容是,从酶蛋白质序列出发,研究酶蛋白质的家族分类及亚类分类(功能预测);酶蛋白质的超二级结构β-β模体(包括β-发夹和β-α-β模体)的分析及预测;酶蛋白质的β-发夹和β-α-β模体功能位点的氨基酸保守性及预测;预测的网页软件原型系统开发。

英文摘要: Enzyme protein is necessary and universal macromolecule for life,research of its structure and function has important relation with development of new drugs, the harvest of the crops and the biochemical synthesis.And experimental methods to study the structure and function of the enzyme protein itself is low throughput,cannot be used to annotate a large number proteins that obtained by developmental genome sequencing technology. Therefore,study and prediction of enzyme structure and function is necessary by using experimental data of high throughput, computer technology, mathematical method and physical model. Enzyme function is often reflected in key binding site, and thus prediction of structure and functional site of enzyme became one of the research hot topics of the protein research. In this project, based on enzyme protein sequences, the research contents include: prediction of classes and subclasses of enzyme;analysis and prediction of super secondary structure motifs β-β (β-hairpin and β-α-β motif); study and prediction of conservative property of amino acid in the functional site; development of webpage software prediction system.
