中文摘要: 本项目以贵州万山汞矿地区长期汞暴露人群为研究对象,采集其补硒后血液、头发样品,通过不同色谱柱并结合电感耦合等离子体质谱(ICP-MS)分离血清、红细胞及头发中汞-硒-蛋白复合物,利用柱后同位素稀释-电感耦合等离子体质谱(ID-ICP-MS)测定其中汞、硒含量,利用电喷雾质谱(ESI-MS)测定其分子量,利用同步辐射装置的X-射线吸收谱(XAS)原位对其结构进行解析,从而实现对长期汞暴露人群补硒后血液、头发中汞-硒-蛋白复合物的鉴定,为阐明人体内硒拮抗汞毒性机制打下基础,同时为利用硒提高长期汞暴露人群健康水平提供理论依据。

英文摘要: In this proposal, the long-term mercury-exposed populations in Wanshan mercury mined area who were supplemented with selenium will be selected and blood and hair samples will be collected. The mercury-selenium-protein complexes in plasma, red blood cells and hair will be seperated using different chromatography columns. The mercury and selenium concentration in these complexes will be studied through isotope-dilution mass spectroscopy and the molecular weight of the comlexes will be studied using electrospray ionization mass spectroscopy. The valence and neighbourhood structure will be studied by synchrotron radiation based X-ray absorption spectrometry. Through all these efforts, this study is aimed to identify the mercury-selenium-protein complexes in blood and hair samples from the long-term mercury-exposed populations. This will help the elucidation of the mechanism of mercury selenium antagnism in human being and provide theoretical basis on the feasibility of the promotion of the health status in long-term mercury-exposed populations through selenium supplementation.
