中文摘要: 本项目针对"罗汉果甜苷Ⅴ#30001;低糖苷结合葡萄糖转化而成,葡萄糖在此转化中具有重要作用,代谢调控增加葡萄糖供应将有利于其积累"的假设,研究发现罗汉果50 d前果实主要含低糖苷Ⅱ、Ⅲ#65292;70 d后果实则仅含高糖苷Ⅴ#65307;品种光合效率高,果实葡萄糖和甜苷Ⅴ#21547;量也高;遮荫降低叶片光合效率,果实葡萄糖和甜苷Ⅴ#21547;量却升高,二者变化呈显著正相关关系;增加叶片光照可提高果实甜苷Ⅴ#21547;量,低温胁迫和抑制呼吸则会降低果实甜苷Ⅴ#21547;量;果实甜苷Ⅴ#24613;剧积累的50 d至70 d,苷元合成关键酶和葡萄糖基转移酶基因表达水平均大幅提高,从而初步揭示罗汉果甜苷Ⅴ#21512;成与葡萄糖代谢相关,但葡萄糖供应量充足,不受生育后期叶片光合效率影响,甜苷Ⅴ#21512;成的主要限制因子可能为相关酶活性,并首次克隆了37个相关酶基因,推导出了其路径模型。这些为罗汉果栽培、育种和采收加工的技术创新提供了理论依据和基因资源。

英文摘要: According to the hypothesis that glucoses played an important role during the mogroside II to mogroside V biotranformation, and improving glucose supply helps to mogroside Ⅴaccumulation. This research found: (1)fruits mainly contained mogroside Ⅱ and Ⅲbefore 50 days, but fruits only contained mogroside Ⅴafter 70 days. (2) The stronger photosynthetic rate of varietis, the higher glucose and mogroside Ⅴcontent of its fruits. (3) Shading reduced photosynthetic rate of leaves, but improved glucose and mogoside Ⅴcontent,there were signifficant positive correlation between them. (4)Increasing illumination could improve mogroside Ⅴcontent of fruits,but low temperature stress and inhibiting respiratin would reduced mogroside Ⅴcontent of fruits. (5) Gene expression level of key enzymes relating to aglycone biosynthesis and gulcosyltranferases greatly rose, as mogroside Ⅴcontent from 50 day to 70 day. The correlations between mogroside Ⅴbiosynthesis and glucose metabolism were revealed. But, the glucose supply were enough for mogroside Ⅴbiosynthesis and not affected by leaf photosynthetic rate during late growth stage. The main limitation of mogroside Ⅴbiosynthesis were related enzymatic activity. Thirty-seven genes of mogroside Ⅴwere cloned. We had also deduced the first putative mogrosides biosynthesis pathway in Siraitia grosvenroii. These will provide theoretical basis or gene resuorces for technological innovation of Siraitia grosvenorii culturing, breeding, harvesting and processing.
