中文摘要: 本项目旨在前人研究基础上,以中国荷斯坦奶牛为研究对象,利用转录组测序、microRNA测序、酵母双杂交试验等高通量技术和生物信息学方法,研究乳腺组织在一个完整泌乳期(从妊娠后期至泌乳结束)内,其分泌细胞从血液中获取脂肪(FA import into cells)、脂肪酸从头合成(de novo synthesis)、外源固醇转运、脂肪酸激活及胞内运送、脂肪酸细胞内合成和去饱和(Fatty acid synthesis and desaturation)至脂滴形成和分泌(Lipid droplet formation and secretion)等代谢过程的转录及转录后调控机制,并在转录水平上探索脂肪生成、代谢调控和乳腺免疫调控的相互关系,为厘清奶牛乳腺细胞脂肪合成、代谢机制,实现乳成份的定向调控选种,生产优质原料奶奠定实验基础。

英文摘要: Based on recent work of other researchers related with fat fat synthesis and metabolism, this program will use transcriptomic sequencing, microRNA sequencing, yeast two-hybrid system and other high throughput technology and bioinformatics methods to explore the regulatory mechanism of fat metabolism of several main processes in the mammary tissues during a complete lactation period, including FA importing into cells, de novo synthesis, Xenobiotic and Cholesterol transport, Acetate and FA activation and intra-cellular transport, Fatty acid synthesis and desaturation, Fatty acid synthesis and desaturation and Lipid droplet formation and secretion. The relationship between synthesis and metabolistic regulation of fats and immunity of mammary was also explored at the level of transcription. The research would pave a good foundation for elucidating the regulation of fat synthesis and metabolistic regulation, and even the components of cow milk to produce high quality raw milk.
