中文摘要: 鞘蕊苏的植物名为毛喉鞘蕊花(Coleus forskohlii),主产印度,我国云南亦稀有分布。民间将该药材用于治疗哮喘、肿瘤等疾患,疗效显著,称其为"神药"。湖北福人药业公司以鞘蕊苏为君药,配伍前胡、甘草组方开发为 "鞘蕊苏胶囊" 新药,并将其移植到湖北通城县规模种植。本项目针对种植鞘蕊苏主要有效成分含量偏低的情况,探讨其生物合成途径的影响因素,以期解决该药材二萜类成分含量富集的科学问题。拟首先构建鞘蕊苏cDNA文库,为萜类合成酶(TPS)基因研究提供平台,然后高通量筛选鞘蕊苏TPS基因,分析其结构特点与功能,阐释其表达特征与分子调控机制,从而为种植鞘蕊苏有效成分isoforskolin含量富集提供科学依据。本项目预期研究成果应用前景广阔,有望提高种植鞘蕊苏药材资源品质,从源头保证"鞘蕊苏胶囊"的临床疗效,同时为提高我国种植中药材有效成分含量积累经验,可望产生良好的经济效益和社会效益。

英文摘要: Coleus forskohlii, mainly distributed in India, and also rarely in Yunnan, China, is extensively used in the treatment of asthma, cancers, etc., which has magical efficacy. The new Chinese medicine Qiaoruisu Capsule which mainly contains C. forskohlii, Radix Peucedani, and Radix Glycyrrhizae was developed by Furen Pharmaceutical Co., Ltd. The C. forskohlii plant was also cultivated in Tongcheng, Hubei Province. This project aimed to solve the problem of low concentration of effective components in C. forskohlii plant, investigate the factors influencing biosynthesis of effective components, and favor to uncover the scientific problem of improving diterpene accumulation in C. forskohlii. The cDNA library will be constructed to facilitate the study on terpene synthase (TPS) in C. forskohlii. Then high throughput screening of the TPS gene will be carried out followed by analyzing structure feature and gene function. Finally, the expression profile and regulation of the TPS gene will be elucidated to provide a scientific basis for the improvement of effective component isoforskolin in C. forskohlii. This study will improve the quality of cultivated C. forskohlii plants, guarantee the efficacy of Qiaoruisu Capsule, thus it will have broad application prospects.
