中文摘要: 许多重要的人类神经退行性疾病都与蛋白质淀粉样纤维在机体组织中的异常聚集有关。本课题拟通过理论分析和数值模拟两方面来研究这一问题,建立定量描述蛋白质淀粉样纤维形成过程的数学模型,讨论不同实验条件对纤维动力学性质的影响,并应用于实际预测中。主要研究内容包括:1)蛋白质淀粉样纤维形成过程的分子动力学模拟;2)基于化学反应动力学理论的纤维形成过程的建模和分析;3)蛋白质浓度、溶液pH 值、温度等不同实验条件对淀粉样纤维形成过程影响的定量研究;4)成纤维蛋白质的分类;5)蛋白质淀粉样纤维晶体结构和动力学性质的预测。本课题的研究对于阐明与蛋白质纤维化有关疾病的致病机理、寻找抑制和分解纤维的可能途径、开发有效的治疗药物都有着重要的理论意义和实践指导价值。

英文摘要: Many human neurodegenerative diseases are associated with the abnormal aggregation of insoluble amyloid fibrous proteins. In current study, we plan to explore the amyloid-fibril system by both numerical simulation and theoretical modeling. A mathematical model for the kinetic process of amyloid fiber formation will be constructed, with quantitative dicussions and predictions on the effects of different experimental conditions. Our main research includes: (1) coarse-grained molecular simulation for the whole process of amyloid fiber formation; (2) mathematical modeling and analysis based on the kinetic theory of chemical reactions; (3) quantitative characterization of the effects of protein concentration, pH value and temperature etc; (4) classification of amyloid proteins; (5) predictions of fiber structure and dynamics. Our study will greatly enhance our current understandings on the mechanisms of amyloid fiber formation and amyloid-related diseases, and also shed light on the depression and decomposition of amyloid fibrils, the development of effective therapy for amyloidosis etc.
