中文摘要: 癌症与阿尔茨海默病(AD)均是威胁人类生命的重大疾病,两者之间存在十分重要的"反向关联性"。研究与AD密切相关的β-淀粉样蛋白(Aβ)与癌细胞的相互作用,将有助于人们深入理解癌症与AD之间的相互关系。本项目拟采用流动注射-扫描电化学显微镜(FI-SECM)联用技术研究Aβ对HeLa细胞的呼吸作用及形貌的影响。本工作以激光拉制的毛细管电极为基材,利用真空溅射镀膜技术和电化学局部绝缘封闭技术自行制备流动注射-超微电极(FI-UME),利用琼脂凝胶包埋法固定HeLa细胞,在生理条件下通过改变Aβ单体的聚集时间获得不同聚集程度的Aβ蛋白,通过FI-UME将Aβ聚集体分微区引入HeLa细胞表面并与之作用,建立用样量少、原位、动态和现场检测蛋白与细胞相互作用研究的新方法。该工作的开展对于生物医学、新型分析仪器开发均具重要意义。

英文摘要: Cancer and Alzheimer's disease (AD) between which there is a very important "reverse correlation" are two major diseases that threaten human's life. To study the interaction between beta-amyloid proteins (Aβ, which is in close relationship with AD) and cancer cells will help us to understand the relationship between cancer and AD in depth.This project intends to study the affect of Aβ to the respiration and morphology of HeLa cells through flow injection-scanning electrochemical microscopy (FI-SECM) technology. In this project, flow injection-ultramicroelectrode (FI-UME) is fabricated based on the laser-drawn capillary electrode as a substrate with the aid of vacuum sputtering and electrochemical partial insulation sealing techniques. HeLa cells are immobilized through the agar gel embedding method. Aβ proteins of different aggregation degree are prepared by changing the monomer aggregation time under physiological conditions. Aβ aggregates are injected into different micro-areas of the surface of HeLa cells through FI-UME to study the interaction. Ultimately, a sampleless, in situ, dynamic and on-site techique will be established to study the interaction between proteins and cells, which will be of great significance to biomedical field and the development of novel analytical instruments.
