中文摘要: 硅酸盐矿物的生物风化是地表发生的最重要的地球化学现象之一,对土壤的形成、地质元素的地球化学循环、矿物的生物淋滤等具有重要意义。本课题以1株具有显著风化硅酸盐矿物功能的细菌新种——宿松伯克霍尔德氏菌(Burkholderia susongensis)菌株L226为供试菌株,以黑云母为供试矿物,采用转座子插入突变、细菌全基因组序列分析、T/A克隆、RT-qPCR、基因敲除与过表达、回复突变等方法,研究菌株L226中与风化黑云母相关的(新)基因(簇),并对其生物学功能进行解析。预期研究结果将为进一步阐明硅酸盐矿物生物风化的分子机制提供理论依据,并为高效矿物风化细菌在农业生产与生物冶金中的应用提供高效菌种资源与技术途径。

英文摘要: Silicate mineral bio-weathering is one of the most important phenomenon occurred at earth’s surface, and plays a fundamental role in soil formation, geochemical cycling of geological elements, mineral bioleaching, and so on. In this study, the new efficient mineral-weathering Burkholderia susongensis strain L226 and biotite are used as the tested bacterium and mineral. The mineral weathering-related genes and their functions are further studied by a series of molecular genetics methods, including transposon insertion mutation, analysis of whole genome sequences of bacteria, T/A clone, real time fluorescent PCR, gene knock out, reverse mutation and over-expression. The excepted results will provide theoretical basis for further understanding of the molecular mechanisms involved in mineral bio-weathering. Meanwhile, it will provide bacterial resources and technical approaches for the utilization of the efficient mineral-weathering bacterium in the agricultural production and bio-hydrometallurgy.
