中文摘要: 本项目针对当前电化学检测系统存在不能单点、并行、快速检测等问题,结合国际研究的最新进展,在已有工作基础上,创新性的提出将棱镜耦合式SPR成像系统用于局部电化学图像检测的新方法。该方法无需使用微电极或扫描探针,通过分析SPR光学信号变化检测局部电化学电流的改变,具有高分辨率、高灵敏度、无标记、并行、快速痕量检测等优点;并行检测点数大于50个,折射率灵敏度达10e-6RIU,空间分辨率达微米级,达到国际先进或领先水平。本项目在建立电化学和SPR理论关系模型的基础上,主要解决高分辨率SPR成像,集成化微流通池制造和低对比度样品图像处理等关键问题,并开展肿瘤细胞的电化学图像检测及肿瘤细胞与细胞外基质的相互作用机理研究,推进电化学检测技术进入空间分辨和分子水平的研究。本项目成果将成为异相反应分析、细胞局部活动监测以及DNA阵列检测的有利工具,在生化分析、临床诊断、药物筛选等多个领域有广泛应用价值。

英文摘要: Traditional electrochemical detection methods suffer from a few key drawbacks such as unable fast parallel detection, being easily disturbed, limited resolution and so on. Based on the latest international research progress and existing research foundation, this project studies on a new localized electrochemical imaging detection method based on prism surface plasmon resonance theory. The method detects variations in local electrochemical current from optical signals arising from SPR without the use of microelectrodes or scanning probe. It enables local electrochemical measurements on the electrode surface in a number of positions simultaneously with high spatial resolution and sensitivity. The technique is noninvasive fast for analysis with parallel detecting points of more than 50, the refractive index detection sensitivity of 10e-6 RIU, and spatial resolution of micron level. In the establishment of electrochemical and SPR theory relation, the project mainly solves the high resolution SPR imaging, integrated microfluidic flow cell manufacturing and low contrast sample image processing. It can be used to detect tumor cell electrochemical image and tumor cell interactions with extracellular matrix environment advancing electrochemical technology into space resolution and molecular level studies.
