中文摘要: 炎症性肠病(inflammatory bowel disease,IBD)是一种慢性肠道炎症性疾病,其发病机制与肠道微生态失衡密切相关,表现为肠道菌群失衡、宿主肠道黏膜免疫功能异常,继之造成肠道粘膜损伤,且PD-1、PI3K及NF-κB信号通路参与其中。故如何重建肠道微生态是临床治疗IBD面临的重大问题。近来,干细胞移植与粪微生态移植术为IBD患者带来了新希望。骨髓间充质干细胞(bone marrow mesenchymal stem cell,BMSCs)用于移植治疗IBD模型,能定植于损伤肠道并发挥治疗作用,但具体的治疗机制尚不明了。因此,本研究在此基础上进一步探讨外源性BMSCs能否影响肠道菌群的种类,及其与宿主的上皮、免疫屏障的相互关系而重建肠道微生态,并深入探讨PI3K信号通路在其中发挥的调控作用。

英文摘要: The pathogenesis of inflammatory bowel disease(IBD), a chronic intestinal IBD, is closely related with intestinal microecological imbalance manifested as intestinal flora imbalance and abnormal intestinal mucosal immune leading to mucosa damage with involvement of PD-1, PI3K, and NF-κB signaling pathways. How to reconstruct the intestinal microecology is thus a major challenge in its clinical treatment.Stem cell transplantation and fecal microbiota transplantation have recently brought a new hope to IBD patients. bone marrow mesenchymal stem cells(BMSC) were used in treatment of an IBD model, which showed that they effectively migrated to the injured intestine and showed an excellent effect on IBD. However, their specific mechanism in treatment of IBD remains unclear. Whether exogenous BMSCs can inflence intestinal flora,its relation with its host epithelial and immune barrier, and the role of PIK3 signal pathway in their regulation were further studied in the present study by reconstructing the intestinal microecology.
