中文摘要: 肺癌是常见的恶性肿瘤,属于最难治的实体瘤之一。据卫生部公布的第三次全国居民死亡原因调查结果显示,过去30年间,肺癌死亡率在我国上升了465%,肺癌已成为我国首位恶性肿瘤死亡原因。而肺癌疗效得不到提高的主要障碍是诊断时病期已晚。由于认识到早期的肿瘤有可能被治愈,肺癌筛查和早期检测相对其他实体瘤(如乳腺癌,宫颈癌,结肠癌和前列腺癌)的生存有利,因此开发肺癌早期诊断工具具有十分重要的意义。在本人多年开发胶体金层析技术和丝线为平台的快速生物分析基础上,提出采用丝线和新型滤纸为载体,结合纳米金(金纳米粒子,金纳米棒等纳米材料)的使用,构建快速可视化同时检测三种肺癌肿瘤标志物的生物传感平台。并结合酶催化信号放大以及电化学检测提高生物传感平台的检测灵敏度,使之达到临床检验的标准。同时将与合作医院协同工作,对病人血样进行直接检测,所得数据将与医院所采用的ELISA,胶体金层析方法测试数据比较。

英文摘要: Lung cancer is the most occurrence malignancy, one of the most difficult to cure solid tumor. According to the data of The Third Time National Survey Results to death reason of citizens released by health department, lung cancer death rate have raised to 465 % in last three decades, and lung cancer has become the first death reason of maglignancy. The barrier to improve its therapeutic effect, is the difficulty to diagnosis lung cancer in its early stage. More and more people have realized that the cancer can be cured in its early stage, and early screening and detection lung cancer seems to be better than other solid tumors (such as breast cancer, cervical carcinoma, colon cancer and prostate cancer). Therefore, it's really important to develop early diagnosis tools for lung cancer.In the project, we will use thread and novel filter paper as substrate, combined with gold nanomaterials(gold nanoparticles,gold nanorod), to construct novel biosensor platform to realize optical simultaneous detection of three lung cancer related biomarkers. Enzyme based catalytic amplification protocols and electrochemical detection will be applied to these platforms to improve their sensitivities. Our final goal is make these biosensor platforms to reach the requirement of clinical standard. Finally, we sill work with cooperation
