中文摘要: 绵羊产羔数是重要的经济性状之一,显著影响绵羊饲养的经济效益。绵羊产羔数属低遗传力(0.1)性状,常规育种方法遗传改良进展缓慢。基因组重测序技术能在全基因组水平上检测与包括产羔数性状在内的重要经济性状的结构变异和差异基因。本项目在前期育种工作建立起的杜湖F2群体的基础上,根据产羔数性状的分化将F2群体分为高、低繁殖羊群,利用全基因组重测序筛查影响产羔数性状的关键基因和结构变异,结合转录组测序筛选出不同繁殖力羊群卵泡期卵巢组织的差异表达基因并对其进行生物学功能验证,从基因组和转录本测序的大规模数据综合分析,综合筛选出产羔数性状候选区域中全基因组结构变异与转录组表达差异的关键基因,并进一步在试验群体和纯种湖羊、小尾寒羊、杜泊羊和甘肃高山细毛羊等不同繁殖力绵羊品种中对其遗传效应进行验证,以期鉴别到影响产羔数性状的关键基因和结构突变并揭示其遗传调控机理。

英文摘要: The litter size in sheep is one of the most important economic traits that affect the economic benefit of sheep husbandry. The litter size in sheep is a low heritability trait and the genetic improvement through conventional breeding is a rather slow process. Whole-genome resequencing provides a possibility to scan structural variations and differential expressed genes related to economic traits include the litter size in sheep at whole genome level. In our previous work we divided the Dorper sheep × Hu sheep F2 population into two subpopulations: high fertility population and low fertility population, the combined use of whole-genome resequencing and transcriptome sequencing in these two sub-populations provide a potential to scan the genome structural variations in the candidate region related to litter size in sheep and identify the differential expressed gene in ovarian during the follicular phase. We will also analyze the association between the candidate genes we identified and litter size in sheep in our experimental population, Hu sheep,Small tail Han sheep,Merimo sheep,Dorper sheep,Gansu alpine fine-wool sheep and so on. We hope to identify the genome structural variation and key differential expressed genes that affect litter size in sheep and detect the genetic mechanisms.
