中文摘要: 非编码RNA(LncRNAs和miRNA)直接或间接调控癌基因或抑癌基因表达,是近年肿瘤研究热点。本课题组在研究肝癌细胞中高表达的长非编码RNA-HULC时,首次发现LncRNA(HULC)可调控miR-613的表达和活性,形成LncRNA-miRNA的肝癌分子调控的机制。HULC表达量与miR-613的表达和活性密切相关,具有明显的负量效关系。然而到目前,miR-613的生物功能和作用靶基因都不明确。我们初步研究已发现miR-613能够缩短肝癌细胞的生长周期,促进细胞增殖,并通过高通量表达芯片检测和生物信息学关联分析,推测它可能是通过ERRB或Src/PTK途径激活细胞内信号转导。我们将在原有研究基础上深入解析HULC/miR-613作用于肝癌的分子网络调控机制,确定miR-613作用的靶基因及其相关信号转导,为临床筛选和评估更有效的诊断标志物和药物靶点提供重要的实验依据。

英文摘要: The non-coding RNA(LncRNAs and miRNA) affect the expression of oncogenes and tumor-suppression genes directly or inderectly, activate or inhibit the intracellualr signal transduction.LncRNA research upon carcinogenesis is hotspot in recent years. On the research upon highly up-regualted in liver cancer(HULC), we first discovered that LncRNA-miRNA interact and formed new molecular network in liver cancer. In our study, HULC up-expression in liver cancer cells can decrease obviously activity and expression of miR-613, it is negative correlation. However, miR-613 biological function and targeted genes have not been clear up to now. We now have found that miR-613 can shorten the cell growth cycle, promote cell proliferation. Furthermore, through the cDNA bioarray and bioinformatics analysis, it has been postulated that it may play its role by the path of ERBB or Src/PTK. We will go on the research work on the basis of our original data, make further the precise molecular mechanism, and determine the target genes and related signal transduction. In future, we wish that we could provide important experiments basis for evaluating the more effective diagnostic biomarkers and therapetic target molecules.
