中文摘要: 哺乳动物的研究结果表明,脂滴包被蛋白在脂肪细胞脂解过程中发挥着重要的调控作用,但其在家禽上的功能机制尚未阐明。本课题组在前期的研究中发现,鸡脂滴包被蛋白基因在脂肪组织中特异性表达,并在高低脂系肉鸡脂肪组织间差异表达,暗示其与鸡脂质代谢密切相关。本项目将在此基础上,利用免疫组化和激光共聚焦技术对鸡脂滴包被蛋白进行亚细胞定位研究;利用基因过表达和RNAi技术分析鸡脂滴包被蛋白基因在脂肪细胞脂解过程中的功能;利用报告基因、凝胶阻滞和定点突变技术揭示鸡脂滴包被蛋白基因的转录调控机制;利用重亚硫酸盐处理和测序技术分析鸡脂滴包被蛋白基因启动子区的DNA甲基化情况。通过这些研究,我们力图阐明鸡脂滴包被蛋白基因在脂质代谢过程中的功能和调控机制。

英文摘要: Studies in mammals have shown that, Perilipin1 play an important role in regulating the process of adipocyte lipolysis, but its function and molecular mechanism in poultry has not been elucidated. Our group in the previous study found that chicken Perilipin1 gene was specifically expressed in adipose tissues and differentially expressed in adipose tissue between fat and lean line broilers, suggesting Perilipin1 also play an important role in regulating the chicken lipid metabolism. On this basis, this project is to locate chicken Perilipin1 using confocal immunohistochemical and laser scanning methods; to analyze the function of chicken Perilipin1 gene in the process of adipocyte lipolysis using gene overexpression and RNAi; to reveal the chicken Perilipin1 gene transcriptional regulatory mechanism using report gene assay, gel shift and site-directed mutagenesis techniques; to analyze the DNA methylation in chicken Perilipin1 gene promoter using sequencing. Through these studies, we will clarify the function and regulatory mechanism of chicken Perilipin1 gene in the lipid metabolism process.
