中文摘要: 神经干细胞(neural stem cells,NSCs)向神经元定向分化是修复脊髓损伤(spinal cord injury,SCI) 的有力手段,明确NSC的定向分化机制是提高SCI疗效的关键。研究证实,miRNA参与了神经系统的发生和细胞分化的调控过程, 但迄今为止,尚未发现与NSCs定向分化相关的miRNA及其所调控的下游靶基因,亦未见到将其应用到SCI治疗中的相关报道。为此,本项目拟通过NSCs定向分化模型筛选及鉴定出与NSCs向神经元定向分化有关的差异miRNA,预测其可能的靶基因并进行功能验证,初步阐明差异miRNA在NSCs定向分化中的作用及其分子机制,进而以筛选出的miRNA为靶点应用于SCI治疗的实验探索,为NSCs分化机制的研究及SCI的治疗提供新的思路和研究手段,具有广阔的应用前景,有望带来可观的社会价值和经济效益。

英文摘要: The directional differentiation of neural stem cells (NSCs) to neuron is an effective way to the therapy of spinal cord injury (SCI). It is very importmant to identify the mechanism of directional differentiation of NSCs. As reported, miRNA participates in the development of neural system and cell differentiation. But the miRNA relative to the directional differentiation of NSCs and the target genes regulated by it are still not reported. Therefore, in our program we will detect the differential miRNA relative to differentiation model of NSCs and predict the possible target genes and validate them. It is the initial study to illuminate the effects and molecule mechanism in the proceeding of the directional differentiation of NSCs to neuron which regulated by miRNA. Then, the miRNA selected may be used as a new target to the therap of SCI. It may bring a new way to the study of the directional differentiation of NSCs and may provide a new method to the therapy of SCI. It must bring about considerable social value and economic returns.
