中文摘要: 硅量子点因其具有尺寸小、激发光谱宽、发射光谱窄、水溶性良好等优点,特别是对生物体的毒性极小,在生物成像领域有着极其广阔的应用前景。将低毒性的硅量子点与磁性材料结合起来,为发展高性能肿瘤靶向荧光/磁共振双模态生物成像探针打开了新的思路。本课题首先选择微波辅助法、一锅法、微乳液法、刻蚀法等方法制备高性能的硅量子点,并利用掺杂、自组装、封装等方法,使之与磁性材料相结合,生成荧光-磁性复合纳米材料;然后,通过表面功能化在其表面连接上针对肿瘤标志物(靶标)的靶向分子,制备出不同类型的荧光/磁共振双模态靶向生物成像探针,并将其应用于动物活体的肿瘤成像分析研究。本课题是无机化学、材料化学、分析化学、合成化学、分子影像、临床诊断等领域的前沿交叉课题,具有创新性和明显的应用价值。该项目的研究成果可以为肿瘤的发病机理与临床诊疗研究提供强有力的技术支撑。

英文摘要: Silicon quantum dots (Si QDs) have attracted much attention in biological imaging field due to their excellent properties such as ultrsmall size, wide excitation spectrum, narrow emission spectrum, high quality water solubility and little toxicity. It can provide a new approach to develop tumor-targeted fluorescence (FL) / magnetic resonance (MR) imaging probes by combining Si QDs with magnetic materials. This research will synthesize high quality Si QDs using microwave-assisted, one-pot, microemulsion and etching methods, respectively. Then the Si QDs will be combined with magnetic materials by doping, self-assembly and capsulation, respectively. Next, various tumor-targeted FL/MR imaging probes will be prepared by the conjunction of targeting molecules corresponding to tumor markers on the surface of the composite of Si QDs and magnetic materials, and they will be employed for tumor-targeted imaging. This research is a frontier and innovative topic in the field of bio-imaging, which intercross the field of inorganic chemistry, material chemistry, analytical chemistry, synthetic chemistry, molecular imaging and clinical diagnosis, etc. The results of this research can provide support for the pathogenesis and therapy study of tumors.
