中文摘要: 血管紧张素转化酶(ACE)是引发人体血压升高的关键酶。获取具有抑制ACE活性的小分子是降低血压的有效途径。然而,人工合成的ACE抑制剂在临床上具有诸多不良反应,因此开发天然来源的ACE抑制剂具有重要意义。本研究拟从海蜇蛋白酶解产物中分离纯化到高活性的ACE抑制肽,并对其序列进行鉴定。在此基础上,利用分子对接和分子动力学模拟对ACE抑制肽与ACE的相互作用进行研究,从而阐明海蜇来源的ACE抑制肽的作用机制。随后,结合前人已报道的ACE抑制肽共同构建多肽结构数据库,利用比较分子场(CoMFA)和比较分子相似性指数分析法(CoMSIA)对抑制肽的三维构效关系进行分析,寻找影响抑制活性的主要因素,并用于指导天然抑制肽的修饰,获得抑制活性更高的ACE多肽类抑制剂。本研究的顺利进行将提供更多种类的肽类ACE抑制剂,并促进对肽类ACE抑制剂的结构-功能关系和催化机理的理解。

英文摘要: Angiotensin converting enzyme (ACE) has been known to be primarily responsible for hypertension. Therefore, obtaining the ACE inhibitor is an effective approach to against hypertension. However, synthetic ACE inhibitors always have side effect, which makes it necessary to explore natural ACE inhibitor. In this project, we plan to obtain some different ACE inhibitory peptides from the protein hydrolysate of Rhopilema esculentum, and identify the sequences of those peptides. Based on the structures of peptides and ACE, molecular docking and molecular dynamics will be performed on ACE-peptide complex and explore the inhibition mechanism of ACE inhibitory peptides. Afterwards, a peptide structure database containing a large number of peptides will be constructed. Further 3D-QSAR (CoMFA and CoMSIA) analysis on these peptides can provide valuable information for the important determinants on inhibition activity and design of new ACE inhibitors possess higher inhibition activity. Our project can provide new ACE inhibitory peptides, and gain a deep understanding on the structure-activity relationship and inhibition mechanism of ACE inhibitory peptides.
