中文摘要: 在前期工作中,我们已证明家蚕翅原基表皮蛋白基因BmWCP4基因受BmPOUM2和BmAbd-A调控(Deng et al., Insect Mol.Biol.,2011; PNAS,2012)。本项目继续以家蚕为研究对象,以受蜕皮激素和保幼激素调控的蛹期专一表达的翅原基表皮蛋白BmWCP4基因为工作模型,采用分子生物学和发育生物学等方法,研究昆虫激素信号传导途径对变态发育调控的分子机理。研究内容包括:1)BmPOUM2 mRNA剪切机理的分析;2)BmPOUM2-S在家蚕生长发育过程中的生理作用;3)核转录因子BmPOUM2与BmAbd-A的相互作用;4)与BmAbd-A相互作用的其它核蛋白因子的鉴定。本研究以翅原基表皮蛋白基因BmWCP4基因表达的激素调控为切入点,对阐释昆虫变态发育分子调控机制这一科学问题具有重要的理论意义。

英文摘要: In previous study, we demonstrated that BmPOUM2 and BmAbd-A were involved in the regulation of wing disc cuticle protein gene BmWCP4 in the silkworm, Bombyx mori(Deng et al., Insect Mol.Biol.,2011; PNAS,2012). In this study, we will continue to study the molecular regulation mechanism of hormonal signal transduction pathway on the expression of BmWCP4 using pupa-specific BmWCP4 as a study model at molecular and developmental levels. The contents of the study include: 1)the analysis of BmPOUM2 mRNA splicing mechanism. 2)the function of BmPOUM2-S during the development of the silkworm. 3)the interaction between the transcription factors BmPOUM2 and BmAbd-A. 4) identification of other transcription factors involved in the interaction with BmAbd-A. The study will focus on the mechanism of hormonal regulation of BmWCP4 and the results derived from the study will have important significance for explaining the mechanism of metamorphosis of insects.
