中文摘要: 口腔种植技术现阶段已获得极大的成功,然而后续的种植体周围炎和少量的无菌性松动仍促使我们进一步探讨种植体周骨代谢。免疫细胞可通过多种机制调节骨代谢,我们前期研究发现,钛离子除本身作用于钙通道相关蛋白引起胞内钙离子浓度变化外,还可能存在其他效应机制影响钙离子内流进而影响T细胞的功能。线粒体在调控T细胞内钙离子浓度有重要的作用,线粒体的运动、线粒体和细胞膜内外电位的改变都影响胞内钙离子浓度的变化。钛离子可能通过其本身及其蛋白复合物所带电荷改变T细胞膜内外及线粒体膜内外的膜电位,影响T细胞内的钙离子交换,进而导致其细胞生物学行为的变化。本课题运用膜片钳技术,免疫组化、酶标仪和细胞行为分析等技术手段,基于膜电位的角度深入了解钛离子对T细胞内第二信使钙浓度的影响机制,从免疫骨学的层面上进一步阐释种植体周骨代谢及探讨可能的调控方式。

英文摘要: Nowadys dental implant has obtained great success in clinic, however peri-implantitis and a small amount of aseptic loosening has prompted us to further investigate the bone metabolism around the dental implant. In our previous study, we have found that titanium ion affected calcium concentration in the T cell by means of chemical interaction, and other mechanism of calcium ions influx caused by the titanium ion may exist and influence the function of T cells. Mitochondria play an important role in the regulation of T cell intracellular calcium concentration. Movement and membrane potential changes of the mitochondria can lead the change of intracellular calcium concentration. Titanium ions may changes the membrane potential of T cells and the mitochondrial membrane due to its physical feature and its protein complex charge, effects of calcium exchange in T cells , all of which may lead to the change of cell biology behavior. In the issue, we will use all available techniques, including the patch clamp technique, the Immuno-histochemistry, enzyme-labeled instrument and cell behavior analysis.The issue will base on membrane potential view to further understand the mechanism of titanium affecting the calcium concentration which is the second messenger in T cell and interpret the possible regulation mode of the peri-implant bone metabolism in immune osteological level.
